(gnl. rnk.), tablo ;_29 cm.Bu çalışmada, Türkiye’nin bazı bölgelerinde (Akdeniz, Ege ve Marmara bölgeleri) yayılış gösteren Boletus cinsi makromantarların morfolojik ve moleküler karakterizasyonu yapılmıştır. Aynı zamanda, Türkiye’nin farklı üniviersitelerindeki fungaryumlardan temin edilen Boletus cinsi makromantar örnekleri de çalışmaya dahil edilmiştir. Toplanan ve temin edilen örneklerin, tür çeşitliliğini belirlemek amacıyla ITS rDNA gen bölgeleri ITS1F-ITS4 primerleri kullanılarak PCR’da çoğaltılmıştır ve elde edilen sekanslar farklı analizlerle (Neighbor-Joining, Maximum Likelihood ve Minimum Evulotion Method) karşılaştırılmıştır. Morfolojik ve moleküler karakterizasyon sonucu, Boletus cinsine ait toplamda 21 takson tespit edilmiştir. Bu taksonlardan 5 tanesi (Boletus pulcherrimus Thiers & Halling, B. pulchrotinctus Alessio, B. rubripes Thiers, B. rubrosanguineus Cheype ve B. xanthopus Klofac & A. Urb.) ülkemiz için yeni kayıt olarak belirlenmiştir. Bu çalışmada tespit edilen taksonlar için geniş bir şekilde deskripsiyonları yapılmış ve kullanışlı bir teşhis anahtarı oluşturulmuştur.In the present study, it was made morphological and molecular characterization of genus Boletus macrofungi grown in some regions (Mediterranean, Aegean and Marmara Regions) of Turkey. At the same time, Boletus macrofungi samples obtained from fungariums in different universities of Turkey were included in this study. ITS rDNA gene regions of collected and supplied samples in order to determine diversity of the species were amplified by using ITS1F-ITS4 primers in PCR and the sequences were compared with different analyzes (Neighbor-Joining, Maximum Likelihood and Minimum Evulotion Method). As a result of morphological and molecular characterization, a total of 21 species belong to genus Boletus were determined. Of these taxa, 5 (Boletus pulcherrimus Thiers & Halling, B. pulchrotinctus Alessio, B. rubripes Thiers, B. rubrosanguineus Cheype ve B. xanthopus Klofac & A. Urb.) were detected as new record for Turkey. And also in the study, descriptions were made and a useful diagnostic key was created for taxa which were identified.Bu çalışma Ç.Ü.     Bu çalışmada, Türkiye’nin Adana ili Çukurova Üniversitesi kampüsünden toplanan Suillus collinitus (Fr.)’un aroma bileşenleri Tepe Boşluğu Katı Faz Mikro Ekstraksiyon Gaz Kromatografisi Kütle Spektrofotometresi (Headspace Solid-Phase Gas Chromatography Mass Spectrometry, HS/SPME/GC/MS) tekniği ile ve 40 ve 70°C olmak üzere iki farklı ekstraksiyon sıcaklık uygulamalarının denenmesi ile belirlenmiştir. Aroma çalışmalarından önce, toplanan mantar örnekleri morfotaksonomik karakterizasyonun yanı sıra, ITS rDNA gen bölgesinin dizi analizleri ile moleküler olarak da tanımlanmıştır. Çalışma sonucunda; 1-octen-3-ol, 3-hexen-1-ol, 2-octene, xylene, butylacetate ve benzaldehyde ana bileşenleri, 40 ve 70°C sıcaklıklarda sırasıyla %75,71-%83,14, %5,59-%6,14, %3,64-%3,99, %3,21-3.53, %2,69-%2,96, %2,36-%2,59 oranlarında tespit edilmiştir. Farklı ekstraksiyon sıcaklıkları denemelerinde ise, 40°C’de 19 bileşik belirlenirken, 70°C’de 16 aromatik bileşik tespit edilmiştir

    A contribution to the knowledge of Amanita coryli (Amanitaceae, Agaricales)

    The authors present descriptions and illustrations of Amanita coryli, rarely featured in the mycological literature, based on molecularly characterised by nrITS sequences specimens from the Balkan Peninsula (Bulgaria and Turkey). The collections studied here suggest that the species is probably not restricted to the presumed host-trees of the genus Corylus and may also occur with some Fagaceae. Further, the analysis of previously released sequences in public databases show that it is a species with a wide distribution in Eurasia, probably much more common than currently known, but likely confused with other members of the section Vaginata

    Biological responses of a non-target aquatic plant (Nasturtium officinale) to the herbicide, tribenuron-methyl

    To assess its response to the herbicide, tribenuron-methyl, samples of Nasturtium officinale were exposed to 0, 0.01, 0.05, 0.1, and 0.5 mg L-1 of tribenuron-methyl for 1, 2, 4 and 7 days. The influence of this herbicide on the relative growth rate, electrolyte leakage, lipid peroxidation, photosynthetic pigmentation, protein content, and performance of anti-oxidant enzymes, such as superoxide dismutase (SOD), catalase, and ascorbate peroxidase (APX), was examined. The results indicated that tribenuron-methyl, applied at 0.5 mg L-1, affected plant growth negatively. It also was determined that chlorophyll a is the most responsive photosynthetic pigment to tribenuron-methyl exposure. Under stress conditions, the anti-oxidant enzymes were up-regulated compared to the control. The SOD activity was significantly stimulated, while the activity of APX was inhibited. A significant correlation was found between lipid peroxidation and SOD activity. The exposure period and herbicide concentration had significant effects on the biological responses against tribenuron-methyl stress. These results may be useful for clarifying the effect of herbicides on non-target aquatic plants

    Ülkemizde kuzu göbeği mantarı olarak bilinen Morchella türleri tüm dünyada sevilerek tüketilenmantarlardan arasında yer alır. Ticari değerinin yüksek olması nedeniyle, ülkemizin değişikbölgelerinden toplanarak, farklı ülkelere yaş ve kuru olarak ihracatı yapılmaktadır. Türkiye coğrafikkonumu ve çok eski yıllardan beri birçok medeniyete ev sahipliği yapması nedeniyle zengin birekolojik çeşitliliğe sahiptir. Bu ekolojik çeşitliliğin içerisinde mantarlarda önemli bir yerkaplamaktadır. Gen kaynaklarımızın korunması, öncelikli çalışma konularından olmalıdır ve buamaca ulaşabilmek için de öncelikle mevcut çeşitliliğimizin tanımlanması gerekmektedir. Sunulançalışmada, Türkiye'nin farklı bölgelerinden 2007-2010 yılları arasında toplanan örneklerin türtanımları yapılmıştır. Arazide GPS koordinatları alınarak fotoğrafları çekilen örnekler, laboratuvaragetirilmiş, mikroskobik ve morfolojik tanılamaları yapılmıştır. Daha sonra kurutulan örneklerde,DNA dizi analizleri gerçekleştirilmiştir. Çalışmada, Türkiye için 5 yeni Morchella kaydı: Morchellafluvialis Clowez, P. Alvarado, M. Becerra, Bilbao & P.A. Moreau, Morchella americana Clowez &C. Matherly, Morchella dunalii Boud., Morchella importuna M. Kuo, O'Donnell & T.J. Volk,Morchella tridentina Bres. sunulmuş; M. fluvialis ile M. americana Esculenta (sarı) grubunda; M.dunalii, M. importuna, ve M. tridentina ise Elata (siyah) grubunda yer almıştırMorchella spp. known as -kuzu göbeği- in our country is one of the consumed mushrooms in thewhole world. It is collected from different regions of Turkey and then exported to different countriesas fresh and dry due to its high commercial value. Turkey has a rich ecological diversity on accountof its geographical location and home to many civilizations for the earliest years. Mushrooms have avery important place in this ecological diversity. The protection of our genetic resources should be thepriority research topic in Turkey and to achieve this goal, it is necessary to define our diversity. In thisstudy, the species description of Morchella samples collected from different region of Turkey during2007-2010 years was performed. They were photographed in their locations and then GPScoordinates for each sample were recorded. After morphologic and microscopic analyses, Morchellasamples were dried and analyzed using DNA sequences. Five new Morchella records for Turkey werepresented in this study: Morchella fluvialis Clowez, P. Alvarado, M. Becerra, Bilbao & P.A. Moreau,Morchella americana Clowez & C.     Chemical Compounds and Biological Activity of Turkish Santolina chamaecyparissus L. Essential Oil by Microwave Assisted Distillation

    Aim of study: The essential oil of S. chamaecyparissus (SEO) was isolated by microwave assisted distillation system to analyze the antioxidant and antimicrobial activities. Area of study: S. chamaecyparissus were collected from Osmaniye province of Turkey in April 2018. Material and method: The chemical constituents and groups (by GC/MS and FTIR spectroscopy, respectively), antioxidant (by DPPH technique) and antimicrobial activities (by agar disc diffusion method) of SEO were investigated. Main results: Twenty aromatic compounds were determined and artemisia ketone (39.83%), camphor (17.65%), beta-phellandrene (8.03%) and beta-bisabolene (7.32%) were the most abundant. IC50 values which were the indicators of antioxidant behavior of SEO, butylated hydroxytoluene (BHT) and gallic acid recorded as 88.301, 0.212 and 0.021 g/L respectively. The antimicrobial activity results showed that all tested microorganisms (Escherichia coli, Streptococcus mutans, Bacillus cereus, Bacillus subtilis, Candida albicans) were highly inhibited. Highlights: SEO could be a good source of monoterpenes especially artemisia ketone. Its herbaceous fragrance may provide a new usage area in men's perfumery

    Antimicrobial, insecticidal and phytotoxic activities of Cotinus coggyria Scop. essential oil (Anacardiaceae)

    WOS: 000342326400007PubMed ID: 24980636The essential oil of Cotinus coggyria Scop.' leaves was found to be rich in alpha-pinene (43.1%), limonene (21.3%) and b-myrcene (8.5%). In the antimicrobial screening, essential oil was notably active on Staphylococcus aureus ATCC 29213, S. aureus ATCC BAA-977, Candida albicans ATCC 14053 and C. parapsilosis ATCC 22019 using the disc diffusion and volatilisation assays. The fumigant assay of the essential oil caused 70% and 100% mortality on the two pest adults of Acanthoscelides obtectus and Tribolium castaneum at 80 mu L L-1 air concentration at 96 h, respectively. In the toxicity assay on weeds, a dose-dependent decrease was observed in the germination and seedling growth of Silybum marianum and Portulaca oleracea. The present results indicated that oil could be suggested as an effective biocontrol agent in various fields
